A single-beam spectrophotometer is a type of spectrophotometer that measures the absorption of light by a sample compared to a reference blank.  In a single-beam spectrophotometer, the sample and the reference (often a solvent or blank solution) are measured sequentially rather than simultaneously.  The instrument consists of a light source that emits UV or visible light, a monochromator to select specific wavelengths of light, a sample holder where the sample is placed, and a detector to measure the intensity of light transmitted through the sample.  The absorbance of the sample at different wavelengths can then be determined based on the ratio of the intensity of the transmitted light through the sample to the intensity of the incident light.   The instrument has basic functions of photometric analysis and quantitative analysis.
